Protocol for ELISA and analysis of Vmax (enzyme kinetics)?

We are planning to do ELISA/ spectroscopy-based study. How to make it for time and dose-dependent assay, like OD values should be taken for every 20secs for 20 mins? Hence, how to calculate Vmax value?
