Why large fluctuations in the LDH determination?

I have large fluctuations in the individual values in the LDH determination (Kit Promega: CytoTox96 Non radioactive Cytotoxicity Assay) of primary neuron cultures. Anyone know if centrifuging the supernatant would help? Thank you for the answers.
