The protein I'm working with is prominently localized to the nucleolus (by immunofluorescence) in prostate cancer cell line experiments. However, intriguingly, when I performed an IHC staining on prostate cancer tissue samples, the same protein was absent within the cancer cells themselves. Surprisingly, while the cancer cells appeared negative for the protein, the stromal cells in the surrounding tissue exhibited a positive staining pattern. When investigating the stromal prostate cancer cell line, I obtained similar results to those observed in the cancer cell line.
