Can you please explain a process for SOS response research and use of SDS PAGE in it. I appreciate your input.
The bacterial response to DNA damage is known as the SOS response.The SOS response is a global response to DNA damage in which the cell cycle is arrested and DNA repair and mutagenesis are induced.

SDS-PAGE is a technique to separate proteins on the basis of their charge/mass ratio. It is used regularly in the frame-work of "proteomic studies". One way to understand its use is: if you give a stimulus that can elicit an SOS response in your subject or experimental model, then you can isolate protein (either total or organellar) and separate the components on a denaturaing gel like SDS-PAGE. You can also go for 2D-PAGE and then Mass spec based analysis to determine the identity of the protein. Further steps may involve cloning the cDNA coding for the protein and expressing it in a suitable mutant to see its function.
In SDS-PAGE molecules are previously denatured (if the case, subunits are separated and all peptides unfolded) and and equally negatively charged by SDS, then the peptides are separated by molecular mass difference.
