I transfected Lamp-1 GFP plasmid to the HEK293 cell line. When I image the cells in TIRF, I see green fluorescence o the cytosol too. Does anyone have any idea that how it can happen?
Lamp-1 is mainly located in the lysosomal membrane. When there is no other reference protein as a cell location, it appears to be in the cytoplasm. It is recommended to observe the fluorescence of lysosomal membrane-labeled proteins at the same time to better locate Lamp-1
Hi Akram, this is an interesting question. I have studies this little bit in the past. The overexpression of fluorescently labelled LAMP-1 can produce so much background during the fluorescence imaging. Typically you may have less than 2% of LAMP-1 in cytosol (or outside of the lysosme area) in normal cells.

this was a very interesting paper. I could not find the vendor for pyrene-benzothiazolium dyes. could you please tell me where I can look at the product?
