Both methods require calibration to correlate the diameter of the zone of inhibition to the MIC of the antibacterial agent. So, why not just do the MIC experiment instead? You can use the broth microdilution assay to measure the MIC in a 96-well plate format, using 2-fold serial dilutions of the agent.

What do you mean by "evaluate"? Do you have an hypothesis? If it's just gee whiz - some activity, it's easier to use well diffusuin.

Further - well diffusion allows introduction of a solution and for some materials (esp. natural extracts) folks use aq. DMSO to solubilize.

if the antibacterial agents are commonly used antibiotics, there will be a test strip such as Etest that you can use to determine the MIC quickly and easily, at least as a screening step.
