A huge bubble forms in the gel during running as you can see in the photo. This was occured after 1 hour it started to run. The bubbles looks like a second line in the gel. Why does that occur and how can i fix it?
It’s perfectly normal and there is nothing that you need to fix. Indeed, you can see from your protein markers that the gel is running normally. The bubbles are actually useful in confirming instantly that you have good electrical contact with the lid terminals when you set up the tank.
I had a similar issue to this before, it was probably caused by your reagents, check your tris-base specifically. This could be one of the problems. Good luck
I recommend that you always prepare the running buffer fresh. You can also check the pH values of the buffers and maybe you check your Tris-glycine. Good luck :)
