Yes, It is needed.You have to process the container at least with the media in which you are going to culture your cells so that it can feel homely.Good Luck!

Thank you sir.....ok. I have done it with sterile PBS as it was written in manual that we can do it with PBS also I have done it for 1 hr. Is it sufficient to do it for 1 hr only? In the manual it was written that you can do for 5 mins to overnight.
