I treat THP1 cells with 5 ng/ml PMA for 48 hrs to differentiate. Recently, after 24hrs of PMA treatment I see black dots in the cytoplasm. These PMA treated cells are not properly responding to LPS. I have attached the pics for the problematic cells. Please suggest the possible causes for this abnormality and how can i troubleshoot it?

I think the black dot problem is not with the cells. Its with the contrast and microscope. You can also observe black dots in the background, hence it's a problem with microscope not cells. Please increase light of the microscope and do try to capture image an an auto white mode..Hope it helps !
Hi Anindhya, it could also be that your cell culture is contaminated.
To me, those black spots look like yeast or fungal cells binding to the cell surface. A contaminated culture might explain the lack of LPS response.
