I'm working with rotavirus infections of cells. Usually when I infect with RRV the cells die very fast (as soon as 24 h.p.i) and then they usually detach one by one and you can observe a clear CPE. MA104 cells which I use for infection, usually get about 80% confluent or more before I infect, which so far has worked out very well. But lately I have observed something strange. The last two times they have detach in flatbeds not one by one. And none of my colleagues have observed this earlier as well. The temperature has been confirmed to be 37 degrees and the flasks (T25 and T75) have worked out well earlier, and the media has been changed. Have anyone experienced something like this earlier? Could it be the Co2 level that is the problem? (We had some problem with the incubator earlier showing accurate 5% CO2 when it actually wasn't). Can anyone help me?
