Hello ! I seeded fibroblasts from ATCC PCS - 201 - 012. But they look round. They don't have an elongated shape And it's been 7 days.
I used medium DMEM with 10% FBS and antibiotics, Fibroblast Basal Medium
What could be wrong?
Hi, let's start from the basic questions:
- are the cells attached to the surface (and still being round) or they are floating freely in the media?
- are the cells alive? (you can simply check it with Trypan blue)
- is your plastic dish cell-culture approved?

I feel there is some type of environmental stress that is causing this problem.
Stress in culture could be due to the following.
1. Check for any type of contamination.
2. Temperature fluctuation in the incubator.
3. Inappropriate gas mixture in the environment.
4. Check the cell culture surface/substratum.
5. The nutrients in the culture media, especially the serum. Have you switched over to a new FBS (brand or lot)?

I can see that the cells are alive because they shine and remain attached, they do not move. The plates that we use in the laboratory are being used for cancer and muscle cells. There is no problem with those cells.
I recently reseeded fibroblasts in DMEM medium, 10% FBS plus growth factors. After 24 hours they looked round but 3 days have passed and they look square.
