I am staining heterochromatin regions in the post-mortem human brain tissue slices using anti-H3K9me3 antibody (1:100). As I counter-stain the nucleus with DAPI, I expect to see the heterochromatin marks inside the nucleus. In some cells I do see the H3K9me3 dots encircling the inner membrane of the nucleus, but in most of the cells the heterochromatic dots are arranged on one side of the cell, just outside the nucleus (Refer to the picture attached). Can anybody help me understand why I am getting this? Is it some non-specific methylated protein getting detected?
