I hope you're doing well. I've encountered an interesting phenomenon in my research and would appreciate your insights. When running gels and staining them with Coomassie, I'm observing specific bands that appear to be overexpressed in 3 out of 10 SHP2 mutants. After identifying these bands as specific proteins through proteomics analysis, I attempted Western blotting with antibodies against these proteins. Surprisingly, I couldn't detect any differences in protein levels among all of the mutants.

I'm curious about what could be causing this discrepancy between the results from Coomassie gels and Western blot analysis. Any ideas or suggestions would be greatly appreciated!
