I am using a compound X (TNF-alpha inhibitor) which turned out to be highly toxic at 10uM and 20uM. In case of IL-8, I can see the dose dependent reduction from 1uM to 10uM to 20uM (mirror of cell death). But in case of IL-6, the level is same for 1uM (non-toxic concentration) and 10uM (highly toxic concentration). However, there is no IL-6 detection at 20uM (also highly toxic). The level of death for 10uM and 20uM is same (more than 95% death). I could not understand that

1) why IL-8 level is not same for 10uM and 20uM which are killing almost all cells?

2) why IL-6 level is same for 1uM and 10uM but different for 20uM while 1uM is non-toxic and 10uM and 20uM are toxic to the same level?

3) If we suppose that similar level of IL-6 at 1uM and 10uM is true due to some reasons then why it is not in the case of IL-8?

I got same profile after repeating 4 times. Please help me I am really confused with this consistently unusual profile. Thank you!
