Primary fimbria epithelial cells were transduced with HPV16 E6/E7 and subsequently with lentiviral hTERT to generate immortal cell FE25. But this cell did not form a colony when grown in soft agar nor produced tumor in NOD-SCID mice.

Based on the description you provided, it appears that your cell line did not acquire additional characteristics that are associated with cell transformation. Immortalized cells are not transformed, therefore, can not form colonies in soft-agar or tumors in NOD-SCID. Introduction of activated H-Ras into immortalized cells could transform them.
