I was trying to break open the E.coli cells which I have grown at 37 degrees Celsius in LB for 8 hr. The cells were stored at -80 degrees Celsius overnight. Next day after doing the French Press, the broken cells suspended in French Press Buffer had an unusually dark grey color which I haven't seen before after doing the French press. What could be the possible reason behind this?
The unbroken cells tend to scatter light better than the debris generated by the French press. Hence the whitish appearance of the suspension before the French press (think of fog, which also makes everything look pale)
it is due to the reflection of light unbroken cells scatter regularly and broken cell cell scatter light irregularly. Basically after French press cell had broken so they they appeared dark.
Thanks Pierre and Ahmad for your reply. But I was wondering why the color was so dark,its much darker than usual. I have done this before and the color gets darker after French Press but this time it was too dark. Could this be due to some contamination or due to cell death?
contamination may also be but the most significant is cell death.
