I have been trying to do immunofluorescence, however, the biggest problem for me is cells are not seeded well in my culture plate. Originally, I was thinking poly-L-lysine which could help foe cell attaching to the plate surface. But then, when i compare my cell with coating and without poly-L-lysine, the cells grow better without this coating. Is there any explanation for than?

I have coated the plate with 2ml of (5mg/50ml)poly-L-lysine for 1hr, then let them to dry. before using it, i washed with PBS.
