I was performing IHC on paraffin embedded mice vascular tissue ,after examining the sections under microscope,I found irregular transparent spots on top of my tissue section, please help,thank you.
Put the slides in xylene for 1 day or more. Remove coverslips and cover again with new mounting.
To remove the coverslips is a good idea. I would recommand to rehydrate the sections. Rinse intensivly , several changes, in distilled water and rehydrate, clear and mount again.
These deposits seam to me a kind of cristals which could come from the last buffer rinse. So before you start dehydrating your slides rinse them in distilled water.
I think these artifacts are due to a bad dehydration (due to short time or hydrated alcohols)
Leave the samples in xylene until the coverslip falls off.
Rehydrate (several changes of descendant graduation alcohols till distilled water) and try to make a better dehydration this time.
