Hello. I´m trying to grow yeasts (S. cerevisiae and C. albicans) in both YEPD and Sabouraud agar, but I don´t see any colonies even after 48 h of incubation at 28 or 37ºC. What could be happening? I´m inoculating the agar plates from liquid cultures in Mueller Hinton broth, were they seem to growh well.

Give them some more time. I have never worked with Candida, but with Pichia and Saccharomyces 48 h is more like the minimum. Although YEPD and Sabouraud are complex media, some nutrients might not be present at concentrations high enough to support vigorous growth (especially when working with auxotrophs).

Definitely incubate at 37. Is the agar plate made by yourself or bought? Is the inoculum fresh? How do you inoculate your plate? Do you use loop or drop on the plate using pastette? I have never had problems with Saccharomyces to be honest and I've been growing them for 6 years. They are normally so easy to grow.
Apparently, the problem was that I wasn't giving it enough time. I waited 24 more hours and the yeasts grew just fine! Thank you all for your answers.
There are many possibilities !!!
First check your media pH which is as per requirement or not.
then incubate your plates at 25oC for 5 days, and also maintain +ve control for same.
Probably because they are growing in liquid medium, when inoculated in solid medium, due to the low concentration of water, there is a drop in its development. Always inoculate young cultures in solid media.
