The flow on macrophages Raw264.7 was conducted. Different from the previous result， the value of SSC-A and FSC-A have increased. Why are SSC-A and FSC-A increased on the flow results of Raw 264.7? Could it be due to cell contamination?
There is a possibility that the gains for these channels were increased. If that is not the case, then the size and granularity of the Raw cells have increased which could be due to contamination or any treatment. We will need more information about the experiment or any treatment protocol involved to make an educated guess.

Tryr cells under a microscope before starting your flow cytometry cell staining. I'm sure you would have seen that they are mostly the same during the experiment. As suggested by Ashutosh Acharya, I'm sure the problem comes from laser gain for the SSC and FSC channel as your cells cannot possibly double in size between the experiments.
