I would like to know the mechanism behind why "membrane impermeable dyes" are not able to penetrate through the phospholipid cell membrane.
Also, does this mean they are also impermeable to regenerated cellulose membranes (for in vitro experiments)?
Specificially looking at the dye called carboxyfluorescein but these dyes in general is OK too!

Usually, it is because they bear one or more charged groups, which makes it very difficult for them to permeate through the hydrophobic core of membranes.
Regenerated cellulose membrane is used for dialysis. Such membranes contain holes through which molecules smaller than the holes may pass. Small molecules like carboxyfluorescein should be able to pass through dialysis membrane, as long as the dye is in a monomeric state and not highly aggregated.
In my experience, some fluorescent dyes do not dialyze as easily as one would expect. My guess is that they tend to form aggregates in aqueous medium, making them too large to pass through the holes as quickly as they should.
Because Membranes have bilayer which consists a hydrophilic part(inside one) and hydrophobic part(outside) Ex. Lipid membrane. most of the dyes are water soluble. Dyes are nothing but very small particles dispersed in water solvent .
Outer hydrophobic part of membranes don't allow water to penetrate and thats why dyes are impermeable to membrane.
Interesting thing is phenomena behind making a surface hydrophilic or hydrophobic.
oil is hydrophobic due to charge differences in water and oil.
Another reason is by varying surface textuers surface can be made hydrophobic. Eg Some leaves.
