Hi, my lab assistant performed an SDS-PAGE electrophoresis as follows: Load order: well 1 corresponds to the standard, well 2-3-4 correspond to BSA proteins, Cytochrome c and Wheat germ acid phosphatase which are 5æl of each protein plus 5æl of water and 5æl of loading buffer were loaded.

Wells 5-6-7 also correspond to the same order of proteins but with 10 μl of each plus 5 μl of loading buffer. (total volume to be charged 20 ul)

Therefore, as you can see, an excess of protein was added and therefore do not consider the concentration of each protein (ug / ul)

The gel used was 10% as it leaves the guide.

I recommend that you discuss about the appropriate range to detect by comassie blue on polyacrylamide gels. They determine the MP of each protein and contrast it with the literature and can also discuss why a large number of bands are seen in the lanes that are presented. Also advantages or disadvantages of loading an excess of proteins.
