Have you faced such a problem?
If yes, please suggest how was the problem rectified.
Its a new equipment, being used for the first time. Under programme block 1, the initial switch time is fixed at 60 secs, final at 120 secs and total run time at 24 hrs. Other parameters are as desired, but even other parameters are fixed.
We are optimising the PFGE work. In the trial 1 we did get results, though would like to optimise a bit more.
We have been using CHEF DRIII for the past 10 years and we have never faced such a problem.
You can adjust all the parameters, nothing is fixed (including the switch times).
I assume you tried editing the parameters before a run is started.
Because the Increase/Decrease keys will automatically get disabled once the program starts.
Did the left display showed “1” while you tried editing the parameters in block 1? If not try pressing the Increase/Decrease keys on the left side of the display panel until "1" is displayed?
If this did not work, try clearing the programs in all the blocks and reprogram it.
FYI, we have successfully optimized PFGE for genome profiling of Vibrio harveyi, Vibrio harveyi-specific phages & clinical isolates of Salmonella and E.coli.

Nothing seems to work. So I have called the Biorad guy and they will be visiting us soon.
