I am wondering if fibroblasts are hard to transfect.

I have been attempting to express a human protein in 293T-cells, but I can not find clear evidence of protein expression in wt or transfected cells by Western Blotting (with several antibodies).

qPCR suggests that there is a large overexpression of transcript of said gene in the transfected cells. The transcript is not detectable in untransfected cells.

However, Western Blotting of wt fibroblasts indicates expression of the protein.

Could it be that the 293T-cells cannot express this protein?

I am thinking about a transient transfection of human fibroblasts using Lipofectamine 2000, but I can't find much information on the topic.

Does anyone have any tips/experience with this?
