Dear Scientists,
I am a Master student and I am doing an anti-cancer effect on LNCaP cell line. I have not had any experience with this cell line before. I found on ATCC and some papers, they recommended using RPMI 1640 1x with 10% Fetal Bovine Serum and 1% Penicillin Streptomycin (I used medium from corning). Unfortunately, they are growing very slow. Does anyone have experience with this cell line? Could you give me some advice on how to take care of them better?

We used to grow our LNCaP cells in RPMI 1640 with 10% FBS and they grew fine, if they have been recently frozen they may take a little time to pick up the pace, also make sure the CO2 concentration in the incubator is 5%, otherwise the pH of the media will rise and impair their growth and/or kill them.

The growth inhibition is usually due to calcitriol in LNCaP cells by creating conditions under which these cells grow very poorly.
My suggestion for a proper oncometabolism to use RPMI 1640 1x with 5% Fetal Bovine Serum (FBS) which will support robust cell growth.

Thank you Dr. Philip G Penketh as you said my cell was frozen and the cell passage is quite high (45). I saw that when the density of the cell was low, they grew slower. Could you let me know your cell density on a 100 mm plate?

We used smaller plates but it would correspond to seeding your plates with around 2 x 10^6 cells.

I agree with Dr. Philip G Penketh RPMI 1640 with 10% FBS will surely work. In the first week, LNCaP generally grows very slowly. Also bear in mind that LNCaP grows in loosely attached clusters.
