I think you want to test a specific sequence that should be inserted as a 3' UTR in a reporter system. In this case pSEAP2 is not going to be the best option. Thermo Fisher scientific has a vector called pMIR-REPORT that uses luciferase and allows insertion of sequences at the 3' UTR. This is the link to their page,

I believe this is a better alternative to test your miRNA target site.
