I want to measure intracellular cAMP in various cells. Which simple method is available? I read some papers about EPAC BRET, but I am looking for a SIMPLE method. I have a decent microscope with UV lamp, filters etc. and a good color camera, all from Zeiss.
In the early 1990ies Roger Tsiens group (Bacskai et al, Science, 1993) used an injectable fluorescent version of cAMP-dependent protein kinase (PKA) to measure intracellular cAMP levels in snail neurons (wich have about 20-50 nM PKA intracelluarly). I tried this in rat pinealocytes (which have more like 1 or more µM PKA in the cytoplasm) for about half a year and couldn`t bring it to work. The reason is probably a phenomenon called "scrambling", means once dissociated this labeled and injected form of PKA re-associates with intrinsic non-labeled PKA subunits and the FRET is gone. You may ask Prof. Carsten Hoffmann in Würzburg for more modern tools for intracellular cAMP measurements, but I am afraid it all needs more than a "normal" fluorescence microscope.

wir haben den Abcam Antikörper benutzt um cAMP erfolgreich zu messen und es sind aber zellspezifisch immer die im Anhang 2 beschriebenen Probleme aufgetreten.

Wir haben natürlich die Bindung des Antikörpers durch DAB Anfärbung visualisiert und dann das ganze mit ImageJ ausgewertet. Die Daten und Befunde wurden aber durch die in dem zweiten Anhang aufgelisteten Probleme verfälscht und daher ist diese Methode nicht unbedingt geeignet cAMP zuverlässig zu messen.Der Hersteller kann sicher heute auch zielführende weitere Hinweise geben. Diese können vielleicht nun eine reproduzierbare und somit gezielte Messung von cAMP ermöglichen. Damals erschienen uns die Ergebnisse der Methode nicht zuverlässig genug. Herzlichen Dank, Forschung ist wirklich nichts für Warmduscher und ein ewiger Kampf gegen die Kalamitäten der eingesetzten Techniken. Hier sind wir deshalb auch immer sehr kritisch. Vielleicht kann man aber doch irgendwie die unspezifische IR und das Quenching in den Griff bekommen. Das ist sichr auch probenabhängig. Ganz liebe Grüße und alles Gute, Burkhard Poeggeler

I also apologize for answering in German and should have communicated in English. The first attachment was on an antibody that allows for the detection and measurement of cAMP and the second attachment describes the general challenges and difficulties associated with these types of assays. They are at best, semi quantitative, and we had a lot of non specific immunoreactivity and quenching. Therefore, we could not reliably measure cAMP. However, now more sophisticated approaches are feasable using the antibody with more robust secondary visualisation and quantification by DAB method to measure cAMP in a simple assay. JImage and other free software can be reliably used to quantifiy the staining by simple microscopy and thus, there may be a chance to come to a sound solution.

The biosensor works very nice and the calculation of ratiometric FRET are less complicated that the other type of FRET measuring.
