I am culturing EOL-1 cell line (a human eosinophil cell line) and I am trying to induce the activation of EOL-1 cells to release ECP by using Leukotriene E4 (300nM). Cells were seeded into serum free media (RPMI 1640) for around 6 hours and then treated with Leukotriene E4 (300nM) dissolved in Ethanol for 30 min, 6 hours, 12 hours and 24 hours. However, when I repeated the experiments at passage 18 and 20, the results were contrast. I don't know if it's due to the passage number or experiment bias? Do you know which passage is optimal for EOL-1 cell activation?
