Make histological cross section, cut sections in microtome and stain them. Then you can use light microscopy to measure the thickness for cuticle wax. Please refer to our paper on Murraya koenigii, which explains that process and procedure.
Mohammad Khodaei Dear sir, i did the SEM by following the protocol and now i have images of cross section, but its little complicated to make sure weather its cuticle waxy layer, as i have attaches the images so you can have a look and if you can help me about the cuticle waxy layer then please send me back after identifying thanks a lot
Alexandra Ivanova i have discussed with microscopy lab, after your answer, about he TEM so they said that it can not do for cuticle wax, as by TEM the cuticle waxy layer will destroy,
Hafiz Muhammad Rizwan, TEM is widely used for study of cell wall and cuticle structure for many decades. Here http://www.plantphysiol.org/content/163/1/5/tab-figures-data or here Article Connecting cutin composition to cuticle ultrastructure and p...
you can find perfect TEM images with explanations. These articles are among the first ones that were found by Google ("fruit cuticle transmission electron microscopy"). For TEM of cuticle you can use any protocol or embedding medium, it will be safe.
