I have never worked before with HUVECs. Can I use high glucose DMEM with 10%FBS and 1%L-glutamine, Penicillin streptomycin and sodium pyruvate? Is it better to use RPMI?
My lab doesn't use DMEM for Endothelial cells of any variety. We use Endothelial Growth Media, with the supplements from Lonza. I use primary murine ECs, not Huvecs. You likely need to grow them on a matrix protein, not cell culture treated plastic in order for them to adhere and proliferate. Collagen 1/4 or fibronectin are common options. I use 2% low bloom gelatin, and EGM 2MV+ 7%FBS and it works great for my cells.
The good media for HUVECs cells is endothelial culture medium (ECM) containing 5% fetal bovine serum (FBS) and 1% endothelial cell growth factor (ECGS).
