For a paper, I need to show that my hESC can differentiate into 3 layer.
I have neurons which represents for ectoderm. I need some cells ( really type is not matter) that represent mesoderm and endoderm.
For this I am quite sure with normal FBS supplemented medium will be sufficient.
My question is since there will be planty of different cell types. Do you know any markers that stain a variety of cell types which are derrived from ectoderm or mesoderm.

For endoderm, sox17 is very good marker. For mesoderm, you can use brahcyury (t/bra) or eomesodermin (eomes). Good luck!
I'm doing this for years now: Sox17, CXCR4 for endodermal cells, and brachyury or hand 1 for mesodermal cells. You could also perform a trilineage differentiation using commercially available cells. Anyway-good look with your experiments!
Thank you for all the valuable answers. However, I was looking for a lineage-specific marker. For example, I widely use nestin for NPC cells. But nestin is not expressed in mature neurons.
For differentiation, I cannot use commercially available cells. My main idea is to prove that my CRISPR modified stem cells can create 3 germ layers. I am working on neuronal linage but to submit a disease model paper I need to show the other 2 lineages too.
I have found 2 easy methods to differentiate them into the define endoderm and mesoderm. But before that, I wanted to find if there are linage specific markers since I can differentiate them nonspecific mature cells by using 10% FBS. That way it could be cheaper for me only if I can find a lineage-specific marker to show the linage in these nonspecifically differentiated cells.
As far as I understand there is not a possible marker protein that is expressed in a whole lineage. So I will differentiate them into define endoderm and mesoderm.

Would you like to use a marker for fully differentiated endoderm (intestine,lung, pancreas...) and/or mesoderm (muscle, blood, bones...) derivatives?
