For monocytes, typically marcophages are not in the blood, they are tissue resident. Irin's response is correct. Cd68 is inside the cell and will require an intracellular staining protocol, thus you can use cd11b, cd14, cd16 alone. There are different subtypes of monocytes which you may want to read up on. F4/80 is not useful for human samples.
Hello Kurma, You do not say what type of macrophage you are needing to phenotype? CD14/MerTK would be a reasonable place to start your multiplex - CD11b/15(pan markers), CD16/32/64(M1), CD115(M2). Have a look at invitrogens immune cell guide for current phenotyping antigen suggestions. Best wishes
