Hi All! I am using ITC to study binding of lectin Concanavalin A to glycopolymer decorated with around 40 saccharide moieties (MW ~ 16 000 Da), which is multivalent-type ligand. Binding of corresponding, free saccharide is 1:1 binding process with Ka = 1.8x10^3 M^-1. My question concerns the concentration, which should be used for data analysis. Should I use the glycopolymer concentration, which is 0.11 mM, or should I regard the concertation to saccharide concentration which is 4.5 mM? In the first case I get Ka = 9.9x10^4 M^-1, while in the second Ka equals 2.5x10^3 M^-1. Thanks for your help!
