I have expressed a transcriptional factor in the Cyanobacterial cell. The target protein is tagged with His-Tag. I would like to do Chip assay (using qRT-PCR). Which Kit is better for this assay? The suggestions are highly appreciated.
Sorry to say this; but you probably have not clearly described what you want to do and why.
From what you say, I understand that you have expressed a His-tagged TF in cyanobacteria and you want to do ChIP for that TF in that cyanobacteria. To do ChIP (for your information, it is "chromatin" immunoprecipitation) you need a pool of chromatin; cyanobacteria doNOT have chromatin. You can simply lyse the x-linked cells in a suitable bacterial lysis buffer like those from Novagen, use a bunch of nucleases to fragment the DNA (I am not sure sonication will be effective) and do conventional Ni-NTA pulldown and do qPCR for select genomic regions.
