Which is the right loading control for cell culture supernatants while running WB?
We are validating proteomics candidates from cell culture supernatants. The culture media was supplemented with serum. Now which loading control is best expressed here?
Bovine serum albumin will be, by far, the most abundant protein in the medium because of supplementation with serum, unless you take steps to remove it. I suppose you could use that as a loading control if you can get an antibody to it.
Hi, the amount of BSA will be independent from what the cells produce by themselves.
If you are looking for a protein secreted by your cells you should take the oberall cell number and the live/dead ratio into account.

It is necessary to cleary difference protein coming from cell and which this from medium. So you have to withdraw amount of medium protein in the final count.
