I am trying to do Microcontact Printing with C2C12 cells and human myoblast; to avoid adherence in regions I do not want, I use Pluronic F-127 for one hour after the protein patterning, then before seeding, I wash 3 times with PBS. However after the first cells adhere to the glass coverslip, my cells do not only adhere to the protein but in other regions where I supposed the Pluronic coating is.
Do you have any recommendations to solve this?

Hi Norma, this is kind of a late response. Pluronic is a hydrophobic / hydrophilic block copolymer. It forms like PEG-like brush layer when it encounters a hydrophobic surface. Is your non-protein regions made of some kind of hydrophobic material? Pluronic prefers hydrophobic surfaces in summary. You can look at a couple of my papers in Biomicrofluidics and ACS Applied Bio Materials for reference using Pluronic F-127.
Jonathan Cheng I am also facing the same issue. In fact I am using hydrophobic PDMS surface to coat the pluronics. I found the cells had variable response on the surface. They looked attached in some areas and looked rounded morphology in other areas. Even if the coating is washed away, I suppose cells should not adhere as PDMS is already hydrophobic. Thanks
