Do commercial kits have an advantage over conventional ultracentrifugation? Given that the sample is comprised of serum or urine rather than cells or cell-media, could UC still be effective, yielding enough high-quality exosomes for downstream experimentation?

Personally I would go first with UC after a centrifugation at 6'000 x g to remove cell debris ansd particles. UC can be at 20'000 x g for 90 min, or 100'000 x g for 12h to pelletize exosomes. You may also consider SEC using for instance qEV to remove the proteins and fractionate partially the extracellular vesicles based on size. Nevertheless this process will not fully separate exosomes from other extra-cellular vesicles that are larger, but you may be able to get some initial data at minimal cost and time.
