Dear researchers,
I don't have any experience with the culture of THP-1 cells. I have found that the optimal confluence to begin a culture is 1x10^6 THP-1 cells/ml, and approximately after 1 week, depending on the company, it expands to 10x10^6. Should I change the media every 2 days? When can I transfer them to a flask? How should I do it, and what should be the desired confluence?
Can somebody enlighten me, please?

THP-1 cells are sensitive to cell density. The recommended seeding density is between 3-7 x 10^5 cells/ml. Generally, 5 x 10^5 cells /ml is used. The culture media used for THP-1 is RPMI 1640 +10% FBS + 50uM 2-mercaptoethanol.
Subculture when cell concentration reaches 8 x 10^5 cells/mL. Do not allow the cell concentration to exceed 1 x 10^6 cells/mL. Medium renewal should be done every 2 to 3 days.
THP-1 cells grow better in conditioned media. Therefore, whenever you passage the cells, leave 1-2ml of old media and add fresh media to the culture.
