I am working with CRC stem cells. I like to know which stem cell culture condition is more precise and accepted? 

1. With or without FBS?

2. With or without growth factors? (like for eg., bFGF, EGF, etc)

3. Regular culture dishes or ultra low/non adherent culture dishes?

As of now, I already have tumorspheres growing from quite a long time in just the regular medium WITH FBS (but WITHOUT any growth factors) and in regular cell culture dishes/flasks (NOT ultra low/non adherent). But of course I still have to subject them to FACS for their "stemness" characterization, and followed by downstream applications.

Is the above method, that I am using, is accepted (especially with respect to publications?) else, please suggest me.
