I want to perform LAMP analysis. According to the procedure betaine should be added. Anyone knows the best betaine for LAMP. I found three from Sigma aldrich.
Betaine (Can no.61962) costs less than 40 EUR/50 g
Betaine (Can no.30056) costs less almost 60 EUR/50 mg so it is alnmost 1000 x more expensive.
does anyone know it the cheaper one suits LAMP?
both are the same compound - trimethylglycine, they differs in terms of purity, but the cheaper one should be pure enough in my opinion, so try it first.
Another question is if you really need betaine. Try first to perform lamp without it. If you have good primers, betaine may be not necessary.
Also, why not to start with commercial kits, it may save a lot of time? Check Novazym Poland company (http://www.novazym.pl/) for different LAMP kits.
I agree with Krzysztof; try your LAMP without betaine. In many cases I have found it makes no difference. I expect it matters more if you have a particularly GC-rich or structured target. There are also other additives you could use to get the same effect, e.g. DMSO, trehalose, etc.
I have also found that my assay doesn't need betaine, but it does depend on the GC content of your target. If your target is GC rich then you may need betaine to aid in the separation of the strands.
