MCF-7 cell line is caspase-3 null that means it doesn't express caspase-3 and hence a suitable model for studying the function of caspase-3 by exogenous expression tagging it with fluorescent protein. Transfection of caspase-3 gfp in MCF-7 would express caspase-3 but what is the time window of it's activation?

Its true that MCF-7 breast cancer cell line do not express cas-3 due to mutation in exon 3. It has also been shown that reconstitution of MCF-7 cells with Cas-3 significantly increases sensitivity towards different chemotoxic substances. However, the time window for the activation will depend on the substance, its dose and also on the expression vector. It is hard to give a general time frame for all substances. You definitely need to titrate your compound in terms of dose as well as duration.

@Prafull kumar, You are very right, sir that it depend on the dose and reagent to be used. What if I use 1 uM of staurosporine to induce apoptosis then can you can me the time window of caspase-3 activation?
