I have a bacterial liquid culture able to produce ethanol. I wanna determine the ethanol amounts in the medium. But I am not sure when is the best time to determine alcohol production under fermentation conditions.
Ethanol production from bacteria was depended on specie of bacteria and you can work normal growth curve of your bacteria to known the best time for production
Hi, take some samples in the fermentation, of course is important at the end of this to measurement final ethanol concentration. With these, you can see if the product is or not associate to biomass, and after you can make a kinetic of process
Ethanol is a growth associated product and max organism produces it mainly during their exponential growth period. So try to find the ethanol produced near around the end of its exponential period.
Ethanol is associated to growth, therefore you need to know the growth kinetic, this growth depend on many factors (pH, T, sugar concentration , medium formulation, etc), there are ethanol fermentations since 8 to 48/72 hours. If your carbon sources concentration is low(less than 100 g/l), the process is fast (10/12 hours), but if yuor concentration is high ( 200-300 g/l) yours process take 48/72 hours. To the first condition you can follow the process every 2 hours, in the other case you can follow the process every 8 hours during the first 24 hours and then every 12 hous until the process finished. All depend on your interest ( to study the kinetic or quantyfing the ethanol produced)

I should study multi factorial conditions and now I understand the process. also I wanna know about the qualification method to detect alcohol simply before doing HPLC for each sample. Does anybody know about it. I know the oxidation of dichromate is a way but I can not find any photo of glass wares apparatus set up for this purpose.
