I am doing in situ zymography to measure MMP activity in heart tissue sections. I am trying to decide between different mounting media w/ DAPI.
1. VectaShield Antifade w/ DAPI (H-1200)
2. VectaShield HardSet Antifade w/ DAPI (H-1500)
3. ProLong Gold Antifade w/ DAPI (P36935)
4. Flouro-Gel II w/ DAPI (#17985-50)
Does anybody have experience using all or multiple of them? Please help :)
My colleague used ProLong Gold Antifade w/ DAPI (P36935), but not sure that he was happy. For others I have no idea. In addition it depends to necessity to have long term slides. If you need your samples only for few days you can cover them with pure dry glycerol. Glycerol should have pH around 7.2-7.6, may be adjusted by PBS in proportion glycerol:PBS - 70:30 or less PBS. But usually pure glycerol is fine and around 7.2 pH itself. To avoid sliding of cover-glass on side surface use transparent nail polisher. It also protect from leakage. Drop small amount of glycerol - 100 uL or so. It should perfectly feet the 22x22 mm cover-glass.
I use this recipe always except for long term storage. Especially it is perfect for plastic multiwell plate staining

I have used VectaShield Antifade w/ DAPI for immunohistology for over 10 years and had no problems! The slides store well for at least a few months too (in the few cases when I went back to look them up). Good luck!
In our laboratory, we routinely use ProLong® Gold Antifade Mountant with DAPI (P36935) when performing fluorescent IHC on mouse knee joint sections that were embedded in paraffin. We have used on it sections that were sectioned ~2 years ago today. In addition, we also use it on fixed cells in vitro grown on either plastic or glass coverslips. Good luck!
We use the DAPI Fluoromount G-Southern Biotech in fixed cells, is a water-mounting medium. We have good results
