HepG2 was fine until last Saturday(third picture), but today they detached and the color turned black...(first and second picture)
Are they dead? or contaminated?
After the subculture, I could see them attached to the bottom of the dish which I found it strange, but it seemed to be growing well so I just ignored it....Would that have caused the problem somehow?
I used low glucose DMEM, and the passage number is 99.

My best guess based on your images is contamination.
I agree with Gisela Buetner.
BTW, If they they are not dead now, they soon will be, as these cells don’t survive floating.
I think the dark appearance may be how their fat globules look under phase once cells are contracted.

I think that the possible explanation is that I changed high glucose DMEM to low glucose DMEM so fast that cells got stressed and died....
Normally, these cell should be grown in MEM (5.5 mM glucose), not DMEM high glucose (22.2 mM glucose). I think the low glucose form of DMEM is 5.5 mM glucose.
