Normally as a result of thermal unfolding the intensity increases and the wavelength of max absorption blue-shifts as Nile Red interacts with the hydrophobic amino-acids. Is it possible that as a result of thermal unfolding of a heme-protein the wavelength blue-shifts with concomitant decrease in intensity? Is it possible that heme Nile Red interaction may be resulting in energy transfer?
