I work with very less volume of sample 10-20 ul, as that is the amount of tears we can collect. Increasing sample volume requires multiple visits by the participants for just one assay. So, is there a way that ELISAs could be done with less than the required sample volume?
You can always try diluting your sample and adding the dilution factor in your calculations. Perhaps whatever you are looking for can be detected this way.
You could get the 96-well plates with smaller wells and decrease the volumes of the other reagents as well. What type of Eilsa is it, because if you are referring to cytokines you could try the BD cytokine kits (for Flow cytometry). These work with 10ul of sample.

It depends on the concentration of analytes in the sample. If abundant, dilute the sample. If not, try using sensitive methods of detection like immunoPCR.

Anouk, I am actually working on Eicosanoids and using the commercially available kits from Cayman.
Agnieszka, I thought of this method but the concentration of eicosanoids is very less, picograms per ml in tears which may lead to the concentration being lower than the limit of detection.
Mike, I have not looked at immunoPCR. Will see if this method might work with eicosanoids.
