Looking for a suitable vessel coating to plate B16-F10 cells that will improve their adherence. Collagen, fibronectin and PDL have been a bust. they will be plated in 96-well plates and exposed to wash buffers containing tween 20. Thanks!
Either fibronectin or collagen should work. Tough to find a better matrix than these. You may simply need to fix prior to washing. However, if you are using multiwell format and directed jets of water against the cells, you'll need to change the approach. I don't know of a matrix that would be foolproof in this circumstance, even with fixation.
Thanks Dwayne, we are fixing them but we still lose a significant enough amount to discount any reliable well-to-well comparison. I've found a publication using hyaluronic acid as a coating but the method is cost prohibitive. Thanks for your input, its much appreciated
