After quantify the amount of single strand breaks, I am trying to identify the sequence of single strand brake site on DNA? Do you know if it's possible? what experiment should I use? Thank you.

I am not sure if it would work, but may be you can try to adopt TUNEL assay (usually used to detect fragmented DNA in apoptotic cells) to label single strand breaks with biotinylated nucleotides and then to enrich DNA-fragments with Strep-beads. The purified DNA can then be sequenced.
