Soil suspended in a buffer shall be centrifuged. Now devide supernatant in two parts, heat one part to boil for about 3 min, cool and assay urease activity in this heat-treated (all enzymes in activated) preparation as a negative control and non-heated soil supernatant as well. The later with show soil urease activity if any.
