There are numerous examples where researchers have optimized the amount of antibodies needed for conjugation with nanoparticles . But there is very less data as to what is the concentration of gold nanoparticles onto which the antibodies are attached.

Whenever you develop a lateral flow assay ( LFA) or any assay for that matter, one of the first things you have to do is to optimize your parameters. Before you can do that, you need to identify your assay substrate which in a LFA would consists of your conjugate pad, sample pad, support, etc. Then you choose your antibodies and the size of the gold nanoparticles AuNPs. All these parameters need to be optimized in accordance with the specific application of your LFA because the nature of the various layers of the LFA could affect how much of the assay components would be needed. The other parameters such as AuNP concentration and the Ab concentration during conjugation also need to be optimized depending on the Abs you will use, the Ag and the AuNP.

Basically to couple Antibody/Antigen with Colloidal particle is based on OD of colloidal Gold.
e.g.: 40 Micro gram Antibody required to coat 1 ml of 1 OD Colloidal Gold.
It also very with Particle size like 20 nm, 40 nm, etc..
Also based on PI of Antibody. lead to selection of Coupling pH.
there are many parameter which are critical for coupling of antibody on nano particles.
